皮克特-施彭格勒反应酶研究进展
摘 要:皮克特-施彭格勒反应酶(Pictet-Spenglerases),简称皮氏酶,是指能够催化Pictet-Spengler反应的一类酶的总称,产生高度立体专一性的Pictet-Spengler反应产物。Pictet-Spengler化学反应能够合成异喹啉类和吲哚类生物碱的骨架结构四氢异喹啉和四氢-beta;-咔琳,在这两类生物碱的化学全合成领域发挥着非常重要的作用。与Pictet-Spengler化学反应相比,皮氏酶具有立体专一性、反应条件温和、产率高等优点,在生物碱类药物的化学酶法和合成生物学法合成中具有重要应用。已发现的几种主要皮氏酶主要来源于植物和细菌,并且发现新的皮氏酶目前该领域最活跃和主要发展方向之一。最近有研究暗示真菌界存在皮氏酶。

Abstract:Pictet-Spenglerasesproducing highly stereospecific Pictet-Spengler reaction products refer to the generalname of a class of enzymes which catalyze Pictet-Spengler reactions. Synthesizingthe skeleton structures of isoquinoline and indole alkaloids, namelytetrahydroisoquinoline and tetrahydro-beta;-caroline, was named Pictet-Spengler reaction,and up to the present day, it has become the most successful chemical totalsynthesis strategies particularly towards those alkaloids. Pictet-Spengleraseshave the virtues of stereospecificity, mild reaction conditions and high yield,thus being of significance applications in the synthesis of alkaloids viachemical enzymatic method and synthetic biology method, compared withPictet-Spengler reaction alone. During this period, several keyPictet-Spenglerases originating from plants and bacteria has been found, andthe direction as to the field of exploring new Pictet-Spenglerases has been oneof the most active and main development. Recently, some research suggested thatthere exist Pictet-Spenglerases in fungi.
