线粒体靶向性铜配合物激活p53诱导肝癌细胞线粒体凋亡作用机制研究【摘 要】癌症发病率和死亡率的升高,使得发展高效抗癌物质成为时代潮流,具有较好抗癌作用和靶向作用的铜配合物成为研究重点。
本论文通过相关实验证明一种含有线粒体靶向基团三苯基膦(TPP)的铜配合物通过激活p53诱导肝癌细胞发生线粒体凋亡而具有抗肝癌效果,并初步确定其有效剂量等范围。
首先通过MTT实验对比铜配合物对不同肝癌细胞(SMMC-7721、Bel-7402、HepG2)的作用效果,筛选出对药物最敏感的细胞株,确定有效的剂量范围。
然后通过研究其对人的正常肝细胞LO2的细胞毒性,即LDH、GOT、GTP实验确定对人体产生毒性作用的最高剂量。
最后根据上述两个研究确定最适合的给药浓度范围,并以此范围做细胞周期实验,以确定药物对细胞周期的阻滞作用。

In this paper, through experiments prove that contains a mitochondrial targeting group, triphenylphosphine (TPP) copper complexes through activation of the p53 induce mitochondrial apoptosis of hepatoma cells with anti hepatoma effect, and the effective dose range was determined preliminarily. Then, by studying the cytotoxicity of LO2, LDH, GOT and GTP in human normal liver cells, the highest dose of toxic action to the human body was studied. 为了改善并提高人类的生活质量,发展高效的抗癌药物已成为当务之急。
